We established a platform to validate the accuracy of the T cell recognition peptides, typically derived from algorithms. In fact, outcomes of peptide prediction are not always compatible with real-world immune responses. This discrepancy arises because peptide prediction algorithms are primarily based on HLA structure and the peptide-HLA, often overlooking the natural process of antigen cross-presentation in dendritic cells. Therefore, this in vitro platform helps bridge this gap by enabling antigen presentation using actual proteins. This allows dendritic cells to activate T cells with the natural selected peptides within the context of antigen presentation, rather than relying solely on peptides selected by artificial algorithms. Our tool offers a physiologically relevant method for validating candidate peptides, enabling the analysis of epitope-specific T cell immune responses. The dominant T cell clones generated by this platform are also valuable for diagnosis and their application in cancer immunotherapy.
